Abstract
L-Pyrrolidone carboxylic acid is prepared by subjecting an aqueous solution containing L-glutamic acid to the action of Pseudomonas cruciviae Gray and Thornton, or the enzyme thereof, and recovering L-pyrrolidone carboxylic acid from the resulting mixture, e.g. by filtration, acidification and extraction with a solvent for the product. The micro-organism may be added as an aqueous suspension, as separated cells, or as cell homogenates. The preferred pH is 6-9, the preferred temperature 20 DEG C.-60 DEG C., and the preferred initial L-glutamic acid concentration 10-100 mg/ml. The enzyme of P. cruciviae Gray and Thornton may be prepared by treating the lyophilised cells with supersonic vibration in the presence of phosphate buffer, centrifuging, treating the supernatant liquid with saturated ammonium sulphate solution, centrifuging after 20 hours, adding ammonium sulphate powder to the supernatant liquid, centrifuging and lyophilising.
